{"id":1036,"date":"2022-03-03T00:00:00","date_gmt":"2022-03-03T00:00:00","guid":{"rendered":"https:\/\/new.www.purdue.edu\/newsroom\/?p=1036"},"modified":"2024-08-20T11:20:16","modified_gmt":"2024-08-20T15:20:16","slug":"new-plant-based-compounds-have-been-created-to-treat-alcohol-use-disorder","status":"publish","type":"post","link":"https:\/\/www.purdue.edu\/newsroom\/2022\/Q1\/new-plant-based-compounds-have-been-created-to-treat-alcohol-use-disorder","title":{"rendered":"New plant-based compounds have been created to treat alcohol use disorder"},"content":{"rendered":"<div class=\"purdue-initial-words-wrap\"><p class=\"purdue-initial-words\">WEST LAFAYETTE, Ind. &mdash;<\/p> \n<p>The 2019 National Survey on Drug Use and Health reported that <a href=\"https:\/\/www.niaaa.nih.gov\/publications\/brochures-and-fact-sheets\/alcohol-facts-and-statistics\" target=\"_blank\" rel=\"noreferrer noopener\">14.5 million people ages 12 and older have alcohol use disorder<\/a>, or AUD. This represents 5.3% of that population, according to the National Institute on Alcohol Abuse and Alcoholism.<\/p>\n<\/div>\n\n\n<p>Researchers in <a href=\"https:\/\/www.pharmacy.purdue.edu\/\" rel=\"noreferrer noopener\" target=\"_blank\">Purdue University\u2019s College of Pharmacy<\/a>, the <a href=\"https:\/\/www.purdue.edu\/discoverypark\/drug-discovery\/\" rel=\"noreferrer noopener\" target=\"_blank\">Purdue Institute for Drug Discovery<\/a> and Washington University in St. Louis are synthesizing compounds to treat people affected by AUD. The research was published in the peer-reviewed journal <a href=\"https:\/\/www.frontiersin.org\/articles\/10.3389\/fphar.2021.764885\/full\" rel=\"noreferrer noopener\" target=\"_blank\">Frontiers in Pharmacology<\/a>.<\/p>\n\n\n\n<p><a href=\"https:\/\/www.pharmacy.purdue.edu\/directory\/rvanrijn\" target=\"_blank\" rel=\"noreferrer noopener\">Richard Van Rijn<\/a>, adjunct associate professor of medicinal chemistry and molecular pharmacology in Purdue\u2019s <a href=\"https:\/\/www.pharmacy.purdue.edu\/\" target=\"_blank\" rel=\"noreferrer noopener\">College of Pharmacy<\/a>, leads the research program with Susruta Majumdar, associate professor of pharmaceutical and administrative sciences in the University of Health and Sciences &amp; Pharmacy in St. Louis. Van Rijn said the compounds are based on alkaloids found in the plant Mitragyna speciosa, or kratom. (Purdue Research Foundation photo\/Steve Martin)<\/figcaption><\/figure>\n<\/div>\n\n\n<p>\u201cWe can synthesize a derivative of the natural-occurring kratom alkaloid speciogynine, which has superior potency to reduce alcohol intake,\u201d Van Rijn said. \u201cWe used mouse models of alcohol use and assessment of adverse effects, including monitoring for seizures or hyperactivity. Our synthesized compound does not display adverse effects observed with other kratom alkaloids, including abuse potential, hyperactivity and seizures. As such, this molecule may have utility in humans in treating alcohol use disorder.\u201d<\/p>\n\n\n\n<p>Alkaloids found within kratom, Van Rijn said, are considered less problematic than synthetic opioids or opium-derived opioids like fentanyl, morphine and oxycodeine.<\/p>\n\n\n\n<p>\u201cThe U.S. Drug Enforcement Agency has not scheduled kratom as a substance of abuse,\u201d he said. \u201cMore research is needed to properly establish this, but one hypothesis for their lower risk for abuse is related to their cellular pharmacology.\u201d<\/p>\n\n\n\n<p>Still, Van Rijn said, several steps are needed to further develop the compound to treat AUD.<\/p>\n\n\n\n<p>\u201cWe will look to reduce any potential off-target effects. Kratom alkaloids may interact with multiple nonopioid receptors, so improving the current molecule series to limit those interactions would make the molecule safer,\u201d Van Rijn said.<\/p>\n\n\n\n<p>\u201cWe also need to conduct studies to determine the half life of the drug, how it is metabolized and how much is getting into the brain.\u201d<\/p>\n\n\n\n<p>A provisional patent application on the research was filed by the co-owner, University of Health Science &amp; Pharmacy in St. Louis.
