Dynamic Proteomic Analysis of the Developing Extracellular Matrix

It is known that the chemical and mechanical properties of the extracellular matrix (ECM) regulate cellular behavior. Throughout development, the ECM undergoes extensive proteomic transformations to facilitate tissue growth; however, the regulatory roles of these changes are largely unknown due to the absence of a global proteomic characterization. Previously, there has been a lack of tools available to effectively study the developing ECM proteome (matrisome). As part of my research, I am developing an experimental design that allows for dynamic proteomic analysis of core and matrisome-associated proteins during normal and disrupted mouse forelimb development. Noncanonical amino acid tagging uses methionine analogs to incorporate functional tags into newly synthesized proteins, providing a mechanism to enrich and identify differentially-regulated proteins by mass spectrometry. This experimental technique will be used to determine fluctuations or disruptions in ECM composition during translational stages of development.
