Regulation of DNA methylation during differentiation and its perturbation in the state of disease

During embryonic development, cellular differentiation is mediated by the interplay of transcription factors and epigenetic modifiers. Amongst the epigenetic factors that modulate the stable repression of genes are the DNA methyltransferases (Dnmts). Despite our growing understanding of the functions of Dnmts, the current methodologies applied for the screening of their active targets suffer from limitations. Here, we aim to develop a high-resolution method to map the activity of Dnmts and identify their direct targets. We also intent to identify the mechanisms by which the regulatory elements of cohorts of genes get specifically methylated during development and if disruption in these mechanisms of repression can be related to the origination of cancer stem cells.
